Pfizer Scientist, Hybridoma in La Jolla, California
BioMedicine Design of Pfizer Inc. has an immediate open for Hybridoma Scientist. This Scientist level position will directly contribute towards our efforts for discovery of monoclonal antibodies using hybridoma and/or B cell technologies. The candidate will support different antibody discovery programs while utilizing a broad range of immunology, molecular biology and cell biology-based techniques. The duties of the job include experimental design and execution, analysis and interpretation of data, and presentation of results to the research team. This position will be exposed to the state of art technology in single cell analysis, and the advanced automation system.
Primary responsibilities include but not limited to:
Track the immune response of experimental animal. Test serum titer using ELISA or FACS based method.
Cell culture and maintenance. Generate hybridoma using electronic based cell fusion. Screen IgG-positive clones to identify target specific hits using high throughput automation system.
Sequence the VH and VL of selected hybridoma, analyze antibody diversity, and choose representative clones for recombinant production.
Transient expression of antibodies in mammalian cells. Small scale purification of recombinant antibodies.
Characterize monoclonal antibodies in their affinity, specificity, liability and biological activity.
BS in biological sciences with over 6 years of relevant antibody-related experience, MS with over 3 years of relevant antibody-related experience.
Strong background in cell culture and sterile technique. Experience in maintenance and expansion of hybridomas.
Molecular cloning experience, such as design of DNA constructs, mutagenesis, and/or library construction.
Experience with various immunoassays, such as ELISA, Western, FACS et al.
Good organizational skills, with scientific rigor and critical thinking.
Demonstrated track record in some of the following areas:
Design of immunization strategies to elicit antibodies in rodents. Familiarity with dosing animals, serum collection and titer test. Experience in transgenic mice is a plus.
Antibody discovery using hybridoma or B cell technology, with experience in cell fusion, hybridoma screening, or B cell isolation and amplification .
Antibody screening using protein ELISA, cell-based ELISA and flow cytometry; experience with liquid handling and automation desired.
Antibody characterization with different immunoassays. Experience in multi-color FACS and IHC is a plus.
Experience of hybridoma VH VL sequencing. Successful experience in single B cell cloning and sequencing is highly desired.
