Pfizer Senior Scientist, Single Cell Sequencing in Cambridge, Massachusetts
We are seeking a motivated and dynamic individual to join the Molecular Profiling Group in Systems Immunology. This individual would support the development, validation, and implementation of single cell sequencing for drug-development in Inflammation and Immunology's portfolio areas. This person will execute technical and laboratory aspects of in-house single-cell analysis projects including the design and execution of laboratory experiments, evaluation of test data, as well the preparation of associated documentation and reports. This person must be technically able to do sample collection and single cell isolation, isolation of RNA or DNA extraction, standard Next Generation Sequencing (NGS) workflows and molecular biology associated with single-cell RNA sequencing. Meticulous attention to detail, proficient reasoning and problem-solving skills in experimental design are needed.
Supports the growth of single-cell analysis technology in drug development by supporting ongoing development, execution, validation and troubleshooting of single-cell assays, including single-cell RNA sequencing, single-cell epigenetic profiling, and antibody based single-cell analysis (CITEseq/REAPseq/TotalSeq/Abseq)
Performs operations of different single-cell analysis workflows; from tissue storage, sample preparation, library construction, data generation, data processing to interpretation of results, ensuring high level of quality and reliability of generated results.
Effective testing and optimization of tissue stabilization, dissociation, and purification protocols.
Effectively optimizing and troubleshooting of single-cell RNA sequencing library preparation.
Application of nucleic acid analysis techniques to monitor library construction quality control.
Implements data processing and analysis workflows, bioinformatics pipelines, and data storage procedures in collaboration with internal and external multidisciplinary teams.
Effectively utilizes available resources as needed to achieve timely execution of projects, works with cross-functional teams (internal and external) to execute plans and deliverables.
Serves as a subject matter resource for single-cell analysis technology, provides guidance on experimental design, method or platform selection, and data management to address a specific need.
Educates other scientists, provides hands-on training, and delivers internal and external presentations.
Learns various single-cell analysis platforms, stays on the top of the scientific and technical innovations, and follows new trends and developments in genomic technologies to uphold the department's competitive advantage.
Drafts experimental plans, validation reports, work instructions and SOPs.
PhD in Biology or other related discipline with 0-3 years of experience or MS with 5+ years of relevant molecular and cellular biology experience
Extensive hands on molecular biology protocol and NGS technology expertise required
Must have at least 3 years of documented hands-on (wet lab) experience with NGS assay development, validation, troubleshooting, and reviewing of sample QC data OR tissue dissociation, cell isolation methods (e.g. FACS), cell culture and basic molecular biology (e.g. PCR, cloning).
Must have in-depth knowledge with different standard NGS platforms, bioinformatics pipelines and NGS data processing and interpretation solutions.
Must have experience in DNA/RNA extraction, nucleic acid quantitation and an understanding of quality control analysis.
1+ year of hands-on single-cell sequencing experience
Previous experience with tissue dissociation protocols, single-cell suspension generation required
Knowledge of single-cell RNA sequencing platforms (10x, inDrop, Drop-Seq, SMARTseq, Rhapsody, or other) greatly preferred.
Bioinformatics skills, including programing in R/python (or any other language), experience with command line, and familiarity with statistical software packages is highly desired.
Experience with computational and statistical approaches for high-dimensional data analysis is a plus.
Ability to access and use a variety of computer software developed both in-house and off-the-shelf.
Occasional crouching, stooping, with frequent bending and twisting of upper body and neck.
Light to moderate lifting and carrying (or otherwise moves) objects including luggage and laptop computer with a maximum lift of 15-20 lbs.
